I believe Uncharted 2: Among Thieves has a good chance. Heck, even Batman: Arkham Asylum/Assassin's Creed II are good contenders.
It's still very early to tell, but I just can hardly believe that a game promoting war (in a certain way) can be a Game of the Year... It doesn't make sense to me. Overall Game of the Year needs to have a good story, good gameplay (which I doubt MW2 will have since it's the same fricking engine since Call of Duty 1, meaning that grenade fests and idiot AIs will be there), good graphics, and much much more. I couldn't care less about multiplayer by the way.
