I think it'll go to Bioshock. The reviews of the game have game of the year written all over them. Plus it's an original IP, unlike most other nominees that are pumping out sequels, like Super Mario Galaxy, Super Smash Bros Brawl, Metroid Prime 3, God of War 2, Halo 3, Call of Duty 4, and Ratchet and Clank. Another game I can see sneaking into the race will be Assasin's Creed.
