Well, for shear numbers it has to be Brain Training or Nintendogs.
As for quality, it really depends on the console. You can't compare a good HD console game to a good Wii game because the definition of 'good' is so different (and that's not a rip on either of them)
For HD consoles, I'd say Gears of War or Eye of Judgement. Gears is really well made, and Eye is amazingly innovative.
For the Wii, I don't know. There haven't been a lot of sequels yet. Wii Sports obviously is the game that everyone is talking about, but I honestly think Red Steel has a chance to be a major success this gen. if they manage to make the sequel any good. If they release a second No More Heroes then that takes the cake, but it's questionable whether they're going to do that.
Oh, and The Conduit is gonna be huge!
Edit: Oh, and if you include Guitar Hero, it wins hands down.
Wii has more 20 million sellers than PS3 has 5 million sellers.
