We don't know what games will come out till the end of the year but if I was to make an early prediction based on what has been seen so far and screens from upcoming releases, then I would say Little Big Planet would have the biggest shot at game of the year.
Simply because of the innovation and sheer creativity involved in designing the title. Of course, there's always a possibility of an unannounced game suddenly appearing and blowing us all away but... if nothing new comes around till the end of the year... I'd rate LBP higher than MGS4, GTA, Fable 2, SSBB, Resistance 2, Killzone 2 etc :)
