The ending is perfect.
Do you honestly think Joel would have sacrificed his surrogate daughter for some great cause like saving the world? Protecting one's family is a natural instinct that comes before general altruism. The fact that Ellie isn't Joel's biological daughter only makes the ending more poignant.
There are a lot of other little brilliant things about the ending as well: one of my favorites it how, when Marlene is sacrificing Ellie's life, she's doing it to save the world. But when she's begging Joel for her own life, it's not to save the world -- it's to save herself. She's just as selfish as Joel is. Everyone in The Last of Us is selfish, even if they're trying to do some grand altruistic thing.
Also, the last playable section when you're running from the hospital and the musical theme that played when Joel lost Sarah kicks in is literally one of the most moving moments I've ever experienced.
If I were in Joel's shoes, I would have done what he did without a second thought.
You spend a large amount of time killing non-infected humans who are more concerned with taking what they can and screwing anyone else. Screw them, save Ellie.

I personally really want Ish to be the main character of the sequel.
Here are my reasons:
1. He has an established tragic backstory (the Sewer colony).
2. He's the moral opposite of Joel. The story could be about how he loses his faith in humanity. It could make for an interesting contrast to the original.
3. He is described as not a particularly great fighter, but sneaky and fast as opposed the the massive, unbelievably strong Joel. This could produce different gameplay scenarios focused less on combat and more on stealth, fleeing, and traps.
4. While it is extremely easy to miss, if you look closely at the drawings, you will discover that Ish isn't white. If there's one problem that Naughty Dog has, it's that all of the main characters in their games are white people (also, all of the major black characters in The Last of Us die.)

Good yes, but it would be great if the sacrifice was there and give the world an oppurtunity to change. If they created a cure and everyone was healed wouldn't that cause something more than give up any hope?

Well, when E3 2016 comes around (and I'm pretty sure we can all guess at this point what the last thing shown at the Sony press conference will be, since TLOU2 has pretty much been confirmed at this point), and Ish is shown to be the main character of the sequel, I am so coming back to this post.
