Maybe, maybe not. Some thought early guesstimates for Uncharted 2 & 3 hitting 5-6 million were too high as well. There's just no way to know right now, especially since it's a new IP, but given the buzz and what Naughty Dog's titles have done this gen, I'd say it's a reasonable guess.

It's a new IP and it's releasing in the last year (or second-to-last) as the PS3's life as the latest Playstation console. 5m is a high estimate. No way it goes over that.
The only IPs that sold 5m on the PS3 are Call of Duty games, GTA4, GT5, Fifa 12, BF3, UC2, MGS4, amd AC2. None of these games were new IPs. And most of them had some multiplayer component and/or had long single-player modes. Probably the only games that don't follow these rules are MGS4 and AC2. (I'm being generous by including MGS4).
It took a lot of those games many years to reach 5m. TLoU won't have many years to sell. Also, TLoU doesn't look like its going to be a long single player game and its multiplayer hasn't been confirmed yet.
It's probably going to do (at best) like other top-tier single-player games and do around 4-5m. (Like Assassin's Creed, God of War 3, etc.) And I'm being very generous with that number since TLoU is going to have a lot less time to sell. 6 & 7m is not realistic at all.
