I think we'll have a Direct in the next 2 weeks (I'm betting the next week). So they definitely need to cover all of that, plus some surprise. I'm expecting a Pokémon surprise. Next gen or something like that.

Honestly with what they currently have announced (and assuming Zelda U is until E3), it isn't enough to sustain a 40 minute video, especially now that DLC for Smash and Splatoon has ended. They're gonna need a lot of new announcements at this point.

You forgot about mynintendo, the Nintendo account, and miitomo.
Nintendo accounts start Next week and the rest is coming Next month.

FExSMT is really not a sure thing, if it comes out in the west, it could come out any time in the second half of 2016.

I didn't even know there was a Direct coming up to be honest. I've been so busy with all these things in my life that I'm trying to take care of. Lol XD
But with that said.
News about Twilight Princess HD, Pokken, and Star Fox should be guaranteed.
A European release date for Fire Emblem Fates should be expected along with one final trailer and amiibo support.
Some updates regarding Genei Ibun Roku #FE.
I'd also like a Bravely Second trailer.
They'll mention the 20th Anniversary of Pokémon and touch up on some things like Pokémon GO, as well as officially announce what we all expect, Pokémon Z.

An announcement of the announcement of NX would be great too. And if we're lucky, they'll cap it off with a trailer for Zelda Wii U. Seems very unlikely though.
