I don't think the pic has anything to do with Punch-out. It's just Reggie getting his body ready.

In a boxing position and a pink training outfit just like Little Mac had in the Smash trailer... I think the conclusion that they are teasing something Punch-Out/Little Mac related really isn't that far fetched.

Just like his Metroid pin meant something last year.

Or like the Skyward Sword music used for the Fils-A-Mech video. People thought it was teasing something Zelda related. Rocco from Mega64 later clarified that he chose the music on a whim and knew absolutely nothing about Nintendo's E3 plans.

Most likely this picture is from whatever skit NOA has recorded to reveal their E3 plans, and is not in any way an indication of what Nintendo is working on. A new Punch-Out game is a possibility, though.

I hope we get to watch Link ride over different terrain this time.

Link would have to appear at E3 before anything! (LoZ U not showing up)

I actually predict that there will be something for Zelda U at E3, but it will be one of those bonus trailers at the end that nobody is expecting.  It definitely will not be in their reveal for tomorrow.

Finally. It's about time!
Where can I buy the tickets for the hype train?

It would be neat, but it would also require a lot of prep work on their part, all while likely delaying the games' release. I'm not sure they think it's worth it.

I don't see why that would be the case. They would just be taking the E3 Demos and putting them all on one app. It wouldn't delay anything.

E3 demos are notoriously unstable: crashes, bugs, and other major technical defects are common, even to the point of occasionally bricking consoles. Simply put, the things are too far from a finished product to be reliable, even though the publisher pulls small teams away specifically to craft these demos.

That's the reason why they limited them to Best Buys, with trained presenters, last year. It would do a lot of harm to release those same demos and have the public experience those defects without the filter of a Nintendo employee to both iron out problems and explaint the situation. Doing what you propose would require them to create demos that are good enough to be released, unsupervised, to the public, and that involves a lot more work.

That must be Reggie.

Nope, too thin.

Look where he is, has obviously been working out.

Reggie takes off his hoodie and he hands out a smartphone. The camera zooms in and we see a brand new trailer before E3. Millions of Little Mac fans become instantly excited then disappointed because its not what they expected. But, TreeTurtle is excited as the reveal is StarTropics III. The game gets an instant GOTY 2015 from every website.

You know what would be hype? Live E3 Demos playable at home. Nintendo would be the kings of good PR for for all of E3.

That would be such a brilliant idea

Would kill their servers and the eshop though. It would be briliant if its a single download with required online and deactivation after E3. It combines all the demos from the floor show. I'd love this.
