On topic, I echo the Plants vs. Zombie talk.  I also had quite a bit of fun with My Life as a Darklord, which takes "tower defense" rather literally.

Mud and Blood 2. First time I've played it I was like.. Well this is kinda lame.. Second game, I did a complete 180 and thought it was an awesome game. Shockingly addictive!

Plants vs. Zombies is awesome (though it takes a while until you get a wider range of plants), but Defense Grid is much more focussed on action and more varied in level design.
