The settings are extremely innacurate and the viewing conditions hide flaws in the picture in store. The best bet would be to read reviews and discussions online, AV(S)Forums is a good place to go.
Pick a good brand and only buy what you need. If you're not going to use 120Hz, 1080p content, internet TV and a load of other features, don't buy a TV that has them, it really drives the price up. Also, quickly look up TV technology (LCD, Plasma, LED) to see which one best suites your viewing conditions.
